How can I get stronger and more agile?

Walking for a few minuites aches my legs, I can't do one push-up and I can't run 100m. I'm fed up with it. At the moment I'm just some nerd that can do any math question thrown at him, but is as strong as an insect. I want to get stronger, more flexible and more agile. What do you suggest for a guy that only works out on PE day which only occurs one day a week.
